Bioinformatics Approach for Metabolism Pathways Curation: Carbohydrate Metabolism and TCA Cycle in the Archaeon Sulfolobus solfataricus P2

Background: Metabolic and genomic informatics integrations in organism-specific databases require comprehensive intensive efforts. PathoLogic, a component of the Pathway Tools software package can create complete Pathway/Genome Databases (PGDBs) from sequence annotation files for any organism. This tool predict metabolic pathways using MetaCyc as reference knowledge base. work aimed to apply bioinformatics approach curate PGDB created Crenarchaeon Sulfolobus solfataricus P2. archaeon grows optimally at 80o C pH 2-4. The genome S. P2 was released 2001. Created PGDBs often need manual curations fill gaps that failed detect.
 Methods: We used Bioinformatics curation carbohydrate metabolism pathway (Entner-Doudoroff “ED”) TCA cycle carried out. Literature search well homology-, orthology- context-based protein function prediction methods were followed this Editors program.
 Results: Curation modified number database by adding extra have not been detected PathoLogic. New such semi-phosphorylated ED new variation added Filling holes (missing enzymes) under study also involved process.
 Conclusion: improved serve base annotations reconstructions other organisms especially closely related Archaea.
